Design: Nanopore library was prepared using a bead-free version of the SQK-LSK109 kit protocol and sequenced on an R9.4.1 rev D flow cell. Basecalling was performed using ONT Guppy in high-accuracy mode and only reads passing the default quality filter were kept.

Study: Nanopore-based assembly of many drosophilid genomes

This work is a collaborative project aiming to broaden the scope of genomic resources for studying evolution in the model group Drosophilidae (fruit flies). Samples were obtained from a variety of sources ranging from inbred laboratory strains to wild samples. DNA was extracted from pools of flies. A Nanopore and Illumina hybrid assembly approach was taken to build the genome assemblies. Initial assemblies were generated from Nanopore sequences, polished with Nanopore reads, and finally with paired-end Illumina reads.
